FOXO4-DRI is built from D-amino acids in reversed sequence, a retro-inverso design intended to preserve side-chain topology while resisting proteolysis. Research has focused on its disruption of the FOXO4-p53 interaction, which promotes nuclear exclusion of p53 and apoptosis in senescent cells while sparing proliferating cells.
Structural work has characterized the disordered p53 transactivation domain as the binding target. Preclinical studies span expanded human chondrocytes, aged Leydig cells, keloid fibroblasts, and vascular models.
History
FOXO4-DRI was designed from research showing that senescent cells depend on a FOXO4-p53 interaction to avoid apoptosis. Interfering with that interaction produced selective clearance of senescent cells in early models.
Subsequent work extended the peptide across tissue-specific senescence models and characterized its molecular target in more detail, alongside reports of context-dependent adverse outcomes.
